A RANDOMIZED PHASE VII STUDY OF ABT--888 IN COMBINATION WITH TEMOZOLOMIDE IN TEMOZOLOMIDE REISISTANT GLIOBLASTOMA (IRB #235086)
A randomized Phase I/II study of ABT-888 in combination with temozolomide in recurrent (temozolomide resistant) glioblastoma (RTOG 0929)
- Histologically proven intracranial glioblastoma or gliosarcoma (Phase II) or any high-grade glioma(Phase I).
- Imaging confirmation of tumor progression or re-growth. Phase II patients must have had 2 adjuvant cycles of temozolomide and may not have more than 2 prior regimens for recurrent disease
- Phase I patients may not have more than 3 prior regimens of treatment
ABT-888 is an orally bioavailable PARP inhibitor that possesses an excellent efficacy and pharmacokinetic profile. ABT-888 enhances the anti-tumor activity of DNA damaging agents including temozolomide, irinotecan, cyclophosphamide, and cisplatin.
- Arm 1: ABT-888 plus temozolomide (days 1-21 of a 28-day cycle) (MTD from phase I)
- Arm 2: ABT-888 plus temozolomide (days 1-5 of a 28-day cycle)
